Through the first two episodes, Dio Brando abuses his step-brother, Jonathon Joestar by destroying his relationships.
Their father does not believe Jonathon when he brings it up, and in fact blames Jonathon for making trouble when he confronts Dio about it.
This culminates in Dio poisoning their father under the guise of bringing him medicine, which Jonathon is finally able to prove.

In part 1, Jonathan gets scolded and sent to bed without dinner by his father, as well as some neglect and some period-typical physical punishment such as getting smacked on his fingers. Dio's father abused his son as well.
Part 2 has a racist attack against a black teenager.
In part 4, the teenage protagonist Josuke gets kicked by his mother and has a gun pointed at him by his grandfather, but it's played for laughs and not implied to be abusive. A young boy also is implied to have grown up neglected and gets killed by a man impersonating his father, though he is brought back later. Two teenage characters also were physically abused by their father in their childhood.
Part 5 has the protagonist abused as a child by his stepfather, as well as neglected by his mother. A father wants to kill his teenage daughter.
Part 7 has the protagonist neglected by his father as a child/young teen after his brother died. Part 8 has a young boy who is forced to crossf

A big part of Part 5 is that the Italian mafia sells drugs, including to children, which plays a large role in a character’s backstory. Dio and his father are alcoholics. Many characters, including a monkey, a 50 year old woman who looks twenty, Italians, and a seventeen year old boy are chainsmokers.

if amputation is a sensitive subject for you, please don't watch this series.

part 1: the main villain has his head separated from his body, but he does not die. he also has his body split open. a deuteragonist is cut in half through the torso and killed.

part 2: the main character of the part, joseph, loses his hand in the finale. another character, stroheim, has his leg amputated, and appears later with prosthetic parts replacing a lot of his body.

part 3: one of the later protagonists, a dog, loses a leg in battle, and another human protagonist loses several fingers. one character is presumed to be killed when attacked by a stand, and only his severed hands remain.

part 4: the main villain has a fetish for hands, and keeps the hands of his victims.

part 5: many arc-relevant cases of amputation, including the protagonist and many antagonists throughout.

Mixed portrayals of police throughout the series. In Part 1, policemen attempt to arrest the villain but are brutally killed by him. In Part 2, the main character calls cops pigs and they are shown abusing their power while harassing a black teenager. In Part 4 the main character's grandfather is a policeman who is portrayed in a positive light. In Part 5 one of the main characters is an ex-cop, but the corruption of the police-force is a point in his backstory.

Not really, unless you count Polpo making a semi-joking statement about how his mind goes blank when he eats or that moment Giorno thought his body was a bed. We can't tell if that was a joke or not because turning into a bed might've been his ability of some sort.

In part 8, there are a lot of implied sex scenes. In part 2, a character peeps in on another character who is naked, getting ready for a bath. In part 3, a little girl is almost sexually assaulted by an orangutan, there is a scene where 2 men become magnetized and stick together, where they accidentally get into multiple inappropriate positions, and there is sexual joke when a character transforms into a little kid, and has his crotch cleaned while he’s taking a bath, this as played as a joke as he “doesn’t know why it feel so good.” In part 4, there is a mention of a criminal who has raped many people, but it’s never shown on-screen. In part 5, there is an implication that a 13 year old boy was sexually assaulted by his teacher, and there is a joke that implies one character was giving another character a b*****b In part 6, the main protagonist mentions that she was caught masturbating, and says she is jealous when she learns about an organism that can have sex with almost anything. In part 7, there is a scene between two woman where one tries to seduce the other, as she puts her crotch on the others face, and a 14 year old was attempted to be raped by 2 separate men on 2 separate occasions.

Some parts of JoJo's Bizarre Adventure do have a happy ending; for instance, Part 4, Part 2, and others. Some do not, but most have a 'good' ending but with one to several character deaths along the way. (Only 3 major characters died in 4, however all but one died in part 6)
