In the first episode, a character buries a dead bug. It then comes to life and speaks to her -- the bug is a mentor figure of sorts to the main characters, but he does not usually appear as a bug. From episode 10 onward, three characters are introduced with winged insects for heads.

very no,
there is a transmasc character but he is shown to be just a very normal polite young man (teen because that's the age group focused on in the show) in fact the anime goes into some of the issues that transmascs face, specifically corrective rape something that is rightfully shown as very wrong and harmful within the anime and something that was specifically done to the character because of him being transmasc which is unfortunately accurate to real life
as a transmasc individual myself while i personally have my issues with this anime transphobia is not one of them and i would argue that this show has a very true to life understanding of transandrophobia/the specific intersection of discrimination that transmascs face, that being said it might not be something to watch if you are one of the many transmasc individuals who has experienced this and might get triggered/experience flashbacks or you're just looking for something lighter to watch that's more focused on trans joy

In episode 3, a character is seen with self-harm scars on their arm.
Additionally, in the latter half of episode 7, there is a theme of using self-harm to avoid other issues. Two characters talk directly about cutting, show scars, and one holds a blade to their arm. In the same episode, they are cut along the injury by an enemy, drawing blood, and accept the wound.
(Some scenes are shown as flashbacks in ep. 8)

technically no, however the main villain of the show is a character that is an ai raised by two humans as their child, while this on its own would be fine some of the ways the ways in which the ai's human parents talk about her when she starts behaving differently may be very familiar to someone who has experienced autism specific abuse, saying things like how she's "not a person she's a robot" and saying that she's "not their child anymore" and mourning the "loss of their child" it should be mentioned that they new from the beginning that she was an ai as they created her, the abuse gets very graphic and i personally found it very triggering

Although fatphobia is not used as comic relief, if this theme is triggering for you please be aware that the major point of the third episode is a character telling a fat girl that they could never be friends because she wouldn't befriend a fat person. This causes the girl to desperately try to lose weight (Most likely starving herself) and dying.
