Rem, as Vash and Nai's caregiver for their first year seems to be a doting parent.
Luida and Brad become parent figures to young Vash later on, there are some debatable issues.
[Spoiler]
They locked Vash (a young kid at that time) up for several days/weeks in the beginning, dehumanzing him and talking openly about getting killing him - only starting to treat him better after he saves their plant. Also, big speciesism talk regarding Vash.
Brad is very unfriendly to Vash in his teenage years, even if it seems to ease into a grumpy, protective brother dynamic.
Vash seems to be happy though, and they're looking out for him, accepting him on the ship and looking after him in his adult years.

There is a serum regulary taken by Wolfwoods to heal his fatal injuries -it implies some sort of reliance and could be interpreted as addiction.
The kids that Conrad experiments on are forced to take several medication/drugs ect.
Otherwise, heavy drinking and smoking onsceeen

Roberto smokes and drinks every episode. Wolfwood is constantly smoking. As of episode 5 this has just been background detail; we have not seen signs of withdrawal etc.

Vash is tied up with rope multiple times in episode 1 (though he demonstrates the ability to easily escape). A character is forcibly strapped to a surgical table in episode 5. Characters are sometimes choked or grabbed during combat.

Technically yes, but not in a literal sense. The the last two episodes Knives metaphorically sexually assaults Vash and the other plants, forcing artificial pregnancy and there are other visuals that suggest it’s a metaphor for sexual assault such and Vash’s “gate opening” and Knive’s weapons looking like sperm. During this process Vash experiences what could only be described as a “mind rape” as he’s stripped of all agency and control over his own body.

In episode 6 at around the 11:50* minute mark, after Wolfwood leaves the orphanage, there’s an EXTREME amount of flashing and pulsating rainbow colors that take up the entire screen. It lasts for more than a minute, and ends around the 13:30* mark. (*General timestamp to account for differences in streaming platforms.)
Near immediately after it’s followed by a scene with flashing red lights. The flashing lights end when the alarm stops sounding. It's not quite as bad at the previous scene, but I would advise anyone who's photosensitive to not look at the screen.

Episode 11 has a lot of pregnancy imagery, which continues into episode 12.
In the first three episodes, the character Rosa is pregnant.
In episode 5, there is an old photo of a character as a baby.
